I finally managed to get the printer working again.
I screwed off the whole heating block etc. once more, flashed the firmware with the 1.1.8 version and reinstalled Cura.
Actually still don't know where the problem was. I assume you are right with the levelling guess. Just happy it works again ...

Unfortunately I've cleaned bed and Nozzle right before the print.
For me as newbie it looks s like the filament is bending upwards towards the Nozzle. However as I made sure there is no filament stuck in the tube and the Nozzle itself is free I can't localize the reason ...

I followed your advice and tried to readjust bed and gantry as well as axchanged the tube and pushed it in until stop.
Now my print has imrpoved a little but is still not back to normal.
The skirt and straight lines were printed better but when it came to support ...
